Insulin Resistance and Secretion Metrics

Fasting blood glucose and serum insulin were estimated using the glucometer method (Accutrend alpha, Roche Diagnostic GMbH, Germany) and rat insulin ELISA kit (Wuhan Fine Biotech Co., Ltd., China), respectively. Systemic insulin resistance was estimated using the homeostasis model assessment index for insulin resistance (HOMA-IR) using the following equation: HOMAIR index=[fasting glucose (mg/dl)×fasting insulin (uU/ml)] / 405) . The triglyceride and glucose (TyG) index, is an important predictor for fatty liver and hepatic insulin resistance. This index was estimated using the following formula TyG index=Ln [ triglycerides(mg/dl)×fasting glucose (mg/dl)/ 2] . To determine insulin sensitivity, the quantitative insulin sensitivity check index (QUICKI)=1/ [log fasting insulin (uU/ml)+log fasting glucose (mg/dl)] was used. To determine the secretory capacity of the pancreas, HOMA-B index was calculated as follows: HOMAB index=20×fasting insulin (uU/ml)/ [fasting glucose (mg/dl)63] (Abo-elmatty et al., 2013 (link); Yoon et al., 2016 (link); Zhang et al., 2017 (link)).

Serum Insulin and HOMA-IR Measurement

Serum insulin was measured using a rat insulin ELISA Kit (Wuhan Fine Biotech Co., Ltd. Wuhan, China) according to the manufacturer’s instructions. The results obtained are expressed as µIU/mL. Further, HOMA-IR was used to identify insulin resistance. HOMA-IR was calculated using the following formula [105 (link),106 (link)].
