Chromium Uptake in Freesia Plants

Concentrations of Cr in roots, stems, leaves, and flowers of freesia plants were determined by taking dry samples (0.5 g) and digested separately for 48 h at 75°C with an HNO3:HClO4 by following the methodology of Singh et al. (2023) (link). Every sample was filtered and diluted to 60 mL with distilled water. The Cr levels in the mentioned parts of freesia were estimated by using an atomic absorption spectrophotometer (AAS-ZEEnit 700P, Analytik Jena, Germany).

Determining Lead Content in Brains

In this study, the brains from the treatment and control groups were collected to measure the Pb contents. Atomic absorption spectrometry (AAS ZEEnit-700P) (Analytik Jena AG, Jena, Germany) was used to detect Pb contents by the method of sample handling, according to previous reports in our laboratory [30 (link)]. In brief, the brain tissue was dried and weighed before pre-digestion using 4 mL HNO3 and 2 mL H2O2 for 30 min at room temperature. Then the sample was transported to the digestion machine of Master 40 microwave oven (Shanghai Sineo Microwave Chemical Technology Co. Ltd., Shanghai, China) for further digestion and analysis.

Evaluating Salinity Tolerance in Rice NILs

Phenotyping of NILs along with parents was carried out in randomized complete block design (RCBD) three replications in hydroponics and micro plot during Kharif (June–September) 2015 and 2016, respectively. To evaluate the salinity tolerance of Saltol NILs (Pusa44 and Sarjoo52) 5 traits were recorded namely salt injury score, root length (cm), shoot length (cm), Na+ and K+ concentration (mM g dw−1) and Na+/K+. Shoot length was measured from the base of the plant to the tip of longest leaf and root length was measured from the base of the plant to the tip of longest root. For seedling score, the seedlings after 14 days exposure to salt stress were scored as per standard evaluation system (SES) score for rice (IRRI, 2013 ). The salt injury score 1 was treated as highly tolerant, 3 was tolerant, 5 was moderately tolerant, 7 was susceptible and 9 was highly susceptible. The concentration of Na+ and K+ ions of plant samples were estimated from di-acid digestion (HNO3:HClO4 3:1) of samples using atomic absorption spectroscopy (AAS-Zeenit 700P, Analytik Jena, Germany) as per the protocol of Miller and Rutzke 2003.

Comprehensive Plant Material Analysis

Roots, stems, and leaves were separated and 50 mg plant material were digested in a microwave oven (MLS Ethos, Germany). To ensure dissolution of aluminosilicates a digestion with 1.6 mL 69 % HNO 3 , 0.6 mL 30 % H 2 O 2 , 0.1 mL 48 % HF, and 1 mL 5 % H 3 BO 3 was chosen. Aluminum concentrations in digests of plant tissue were determined with ICP-MS 7700X (Agilent Technologies, Germany). Calcium concentrations were determined with AAS Zeenit 700P (Analytik Jena, Germany). Total nitrogen and total organic carbon (TOC) in nutrient solution were analyzed with a Vario TOC Cube (Elementar Analysensysteme, Germany). NH + 4 -N, NO - 3 -N, and PO 3- 4 -P concentrations in organic layer leachate before and after weekly treatment were analyzed with a Continuous Flow Analyzer (CFA AutoAnalyzer 3 HR , SEAL Analytical, Germany).
