After obtaining sections of aortic sinuses, slides were air-dried for 30 minutes then dehydrated using a sequence of 50%, 95% and 100% ethanol solution, and 100% xylene, and then air-dried again. Lesion and non-lesion portions of the sections were independently microdissected using an Arcturus Pixcell II laser capture microdissection instrument by following the instructions supplied with the instrument. Separate parameters were used for each type of sample removed under arid conditions. For lesion-areas, the following parameters were used: a 15 um spot size, a power setting of 80mW, and 3.0 ms duration. For non-lesion-areas, the parameters set were: a 7.5 um spot size, a power setting of 45mW, and duration of 1.5 ms. Samples were captured on Arcturus Capsure Macro LCM caps and placed into Eppendorf tubes containing 50μL RNA extraction buffer (PicoPure RNA Isolation Kit, Arcturus).

Glomerular and interstitial tissue in the immediate vicinity of the glomerulus was laser-capture microdissection from FFPE sections from patients with IgAN, MN, and TMN (Table 2). FFPE sections (4 μM) were deparaffinized using 3 × 5 min incubations in xylene. Laser-capture microdissection was performed using Arcturus Capsure Macro LCM (Arcturus Engineering Inc., Mountain View, CA) caps on an Arcturus PixCell II.

Laser‐capture microdissection (LCM) was performed using an Arcturus Pix Cell II as previously described.35 Using human prostate cancer tissues and matched adjacent normal tissues, microdissected cancerous and normal epithelial cells were identified from a total of 31 cases by a pathologist in the LCM core facility (National Cancer Institute). Approximately 10,000–15,000 laser shots were used for each case to procure the epithelial samples.33

Laser capture microdissection (LCM) was performed to selectively extract total RNA from OSCC specimens. Tissue sections (7-mm) were prepared from each paraffin block and stained with hematoxylin and eosin. Slides were transferred for microdissection using a Pix Cell II laser capture microscope (Arcturus, Mountain View, CA, USA) according to the manufacturer's instructions. Approximately 5,000 tumor cells were microdissected from each sample. Total RNA was extracted using TRIzol reagent (Invitrogen, Carlsbad, CA, USA).
